Evotec is investing in the strategy, to transfer progressively Evotec’s collection compounds into acoustic tubes (AT) to support our partner’s screening needs. Compounds will still be distributed from Master stock plates for Assay Validation (AV) and Primary Screen (PS) steps. However, for the Hit Confirmation (HC) and Hit Profiling (HP) phases, compounds will be prepared directly from AT.

Enhancing throughput while reducing footprint is a recurring requirement in sample management. With the new Smartcel-2 from Flexible Lab Solutions, we have significantly boosted our capability to prepare assay-ready plates in a serial dilution format. This double-deck automation system features a Bravo from Agilent for high-volume serial dilution plate preparation and a mosquito LV from SPT Labtech for nanoliter dispensing steps. Several peripheral devices, including a peeler, sealer, centrifuge, and plate storage, enable the automation cell to operate around the clock.

With the growing interest in Biologics within the drug discovery field, the Evotec Sample Management teams have been asked to offer their platforms and expertise to enhance throughput and timelines by establishing robust, high-quality processes in aqueous solvents. While extensive experience with DMSO-based solvents was leveraged, the significantly different properties of the water-based solvents raised new questions regarding the handling of source and destination plates at Sample Management.
 

To address these challenges, comprehensive tests were conducted, focusing particularly on evaporation and the establishment of a device verification process. This poster presents these processes and provides an overview of the results.

Biologics manufacturing typically uses engineered Chinese Hamster Ovary (CHO) cells to produce folded and glycosylated antibodies. Determining the optimum conditions to grow and maintain cell culture often requires considerable time and effort.

A quantitative understanding of cell metabolism through an analysis of cell culture metabolites can enable optimization growth conditions for improved titles or increased perfusion duration. Mass spectrometry is the optimum tool for metabolite measurement, however, transforming raw data into accurate quantitative measurement requires both expertise and extensive sample preparation.

In this poster we demonstrate the ability of simple sample preparation using universal calibrators and a novel machine learning algorithm to rapidly provide biological insight into bioprocessing samples taken from perfusion cell cultures.
